产品介绍:This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class I of the sirtuin family. Alternative splicing results in multiple transcript variants.

NAD-dependent protein deacetylase, which regulates processes such as apoptosis and muscle differentiation by deacetylating key proteins. Deacetylates 'Lys-382' of p53/TP53 and impairs its ability to induce proapoptotic program and modulate cell senescence. Deacetylates TAF1B and thereby represses rDNA transcription by the RNA polymerase I. Deacetylates 'Lys-266' of SUV39H1, leading to its activation. Deacetylates 'Lys-26' of HIST1H1E. Involved in HES1- and HEY2-mediated transcriptional repression. Inhibits skeletal muscle differentiation by deacetylating PCAF and MYOD1. May serve as a sensor of the cytosolic ratio of NAD(+)/NADH, which is essential in skeletal muscle cell differentiation. Deacetylates 'Lys-16' of histone H4 (in vitro). Component of the eNoSC (energy-dependent nucleolar silencing) complex, a complex that mediates silencing of rDNA in response to intracellular energy status and acts by recruiting histone-modifying enzymes. The eNoSC complex is able to sense the energy status of cell: upon glucose starvation, elevation of NAD(+)/NADP(+) ratio activates SIRT1, leading to histone H3 deacetylation followed by dimethylation of H3 at 'Lys-9' (H3K9me2) by SUV39H1 and the formation of silent chromatin in the rDNA locus. 免疫荧光技术的实验步骤：

一、准备好试剂与仪器：

磷酸盐缓冲盐水、荧光标记的抗体溶液、缓冲甘油、搪瓷桶三只、有盖搪瓷盒一只、荧光显微镜、玻片架、滤纸、37℃温箱等。

二、实验步骤

1.滴加0.01mol/L，pH7.4的PBS于待检标本片上，十分钟后弃去，使标本保持一定湿度。

2.滴加适当稀释的荧光标记的抗体溶液，使其覆盖标本，置于有盖搪瓷盒内，保温一定时间以三十分钟为参考。

3.取出玻片，置玻片架上，先用0.01mol/L，pH7.4的PBS冲洗后，再按顺序过0.01mol/L，pH7.4的PBS三缸浸泡，每缸三到五分钟，并不停地摇晃振荡。

4.取出玻片，用滤纸吸去多余水分，但不使标本干燥，加一滴缓冲甘油，以盖玻片覆盖。

5.立即用荧光显微镜观察。观察标本的特异性荧光强度。

抗体的定义：

抗体（antibody），（免疫球蛋白不仅仅只是抗体）是一种由浆细胞（效应B细胞）分泌，被免疫系统用来鉴别与中和外来物质如细菌、病毒等的大型Y形蛋白质，仅被发现存在于脊椎动物的血液等体液中，及其B细胞的细胞膜表面。抗体能识别特定外来物的一个特征，该外来目标被称为抗原。

抗体的结构：

抗体是具有4条多肽链的对称结构，其中2条较长、相对分子量较大的相同的重链(H链)；2条较短、相对分子量较小的相同的轻链(L链)。链间由二硫键和非共价键联结形成一个由4条多肽链构成的单体分子。轻链有κ和λ两种，重链有μ、δ、γ、ε和α五种。 整个抗体分子可分为恒定区和可变区两部分。在给定的物种中，不同抗体分子的恒定区都具有相同的或几乎相同的氨基酸序列。可变区位于"Y"的两臂末端。在可变区内有一小部分氨基酸残基变化特别强烈，这些氨基酸的残基组成和排列顺序更易发生变异区域称高变区。高变区位于分子表面，最多由17个氨基酸残基构成，少则只有2 ～ 3个。高变区氨基酸序列决定了该抗体结合抗原抗原的特异性。一个抗体分子上的两个抗原结合部位是相同的，位于两臂末端称抗原结合片段(antigen-binding fragment, Fab)。"Y"的柄部称结晶片段(crystalline fragment,FC)，糖结合在FC 上。
